In this review, (1) we will provide an overview on the precise molecular and ion channel organization of myelinated axons and the reciprocal axo-myelin interactions that warrant properly balanced physiological activities within the CNS, (2) we will dissect the underlying cause of demyelination and the structural and functional consequences of demyelination in axons by focusing on spinal cord injury (SCI) and multiple sclerosis (MS) models, (3) we will discuss the role of activated glia in demyelination and remyelination following demyelination, and (4) we will review the current status of cell-based therapeutic interventions that are designed to promote oligodendrocyte differentiation and facilitate remyelination
